A feasible method to extract DNA from the cambium of high-canopy trees: from harvest to assessment Acta Amazonica
MANGARAVITE,Érica; TERRA,Vanessa; HATTORI,Eric Koiti Okiyama; DAL’SASSO,Thaís Carolina da Silva; BHERING,Leonardo Lopes; OLIVEIRA,Luiz Orlando de.
ABSTRACT Many tropical trees have high canopies and their leaves are not accessible. Thus, the use of tissue from a more accessible organ (cambium) for DNA extraction may be an alternative for molecular studies. We adapted a feasible methodology for extracting genomic DNA from cambium tissue harvested in the field for the assessment with PCR. We tested three storage conditions (two buffers and a silica gel) and four periods of time after harvest. We used previously described protocols and tested them on three species that occur in Amazonian forests and other biomes: Anadenanthera peregrina var. peregrina, Cedrela fissilis, and Ceiba speciosa. Our protocol obtained suitable PCR-grade genomic DNA for DNA sequencing and microsatellite genotyping. We recommend...

Effects of aroma quality on the biotransformation of natural 2-phenylethanol produced using ascorbic acid Electron. J. Biotechnol.
Tian,Xun; Ye,Rui; Wang,Jingwen; Chen,Yifei; Cai,Baoguo; Guan,Shimin; Rong,Shaofeng; Li,Qianqian.
Background Natural 2-phenylethanol (2-PE) is an important flavoring that emits the aroma of roses. During biotransformation, the aroma quality of natural 2-PE is affected by its main by-products, which include butanol, isobutyric acid, butyric acid, and isovaleric acid. Thus, controlling undesirable by-product formation can reduce the effect of odor on 2-PE aroma quality. Results 2-PE was produced through biotransformation using l-phenylalanine as a substrate and glucose as a carbon source. Ascorbic acid was added to the system to improve the redox reaction and suppress the generation of by-products. Principal component analysis of the aroma quality of 2-PE was performed using an electronic nose. Similarity analysis revealed that the effects of four...

Effects of N-acetylcysteine on spinal cord oxidative stress biomarkers in rats with neuropathic pain BJMBR
Horst,A.; de Souza,J.A.; Santos,M.C.Q.; Riffel,A.P.K.; Kolberg,C.; Partata,W.A..
N-acetylcysteine (NAC) inhibits nociceptive transmission. This effect has been associated partly with its antioxidant properties. However, the effect of NAC on the levels of lipid hydroperoxides (a pro-oxidant marker), content of ascorbic acid (a key antioxidant molecule of nervous tissue) and total antioxidant capacity (TAC) is unknown. Thus, our study assessed these parameters in the lumbosacral spinal cord of rats with chronic constriction injury (CCI) of the sciatic nerve, one of the most commonly employed animal models of neuropathic pain. Thirty-six male Wistar rats weighing 200–300 g were equally divided into the following groups: Naive (rats did not undergo surgical manipulation); Sham (rats in which all surgical procedures involved in CCI were...
